本專利技術涉及對SlP受體具有親合性的新型螺-環胺雜環衍生物、含所述化合物的藥物組合物、以及所述化合物用于制備治療、緩解或預防其中涉及任何SlP受體或其中涉及通過任何SlP受體調節內源性SlP信號系統的疾病和病癥的藥物的用途。
技術介紹
1-磷酸-鞘氨醇(SlP)為介導多種細胞反應,如增殖、細胞骨架組織和遷移、粘連-和緊密連接裝配、以及形態發生 的生物活性鞘脂類。SlP可與位于質膜的G蛋白-偶合的受體的內皮細胞分化基因族(EDG受體)的成員結合。迄今為止,該族的5個成員已經被鑒定為不同細胞類型中的 SlP 受體,SlPl (EDG-1)、S1P2 (EDG-5)、S1P3 (EDG-3)、S1P4(EDG-6)和S1P5 (EDG-8)。在許多細胞類型中SlP可產生細胞骨架再排列以調節免疫細胞運輸、血管內平衡以及中樞神經系統(CNS)和外周器官系統中的細胞通訊。眾所周知SlP由血管內皮分泌并以200-900納摩爾的濃度存在于血液中,且被白蛋白及其它血漿蛋白結合。這既提供了細胞外液中的穩定儲庫又能有效釋放到高親合性細胞表面受體。SlP以低納摩爾親合性與5個受體S1P1-5結合。此外,血小板還含有SlP并可在局部釋放引起例如血管收縮。該受體亞型SlPl、S1P2和S1P3被廣泛表達并代表心血管系統中的顯性受體。另外,SlPl還是淋巴細胞上的受體。S1P4受體幾乎只在造血系統和淋巴系統內。S1P5主要(盡管不只是)在中樞神經系統中表達。S1P5的表達似乎局限于小鼠中的少突神經膠質細胞、腦的髓鞘生成細胞(myelinating cells),但在大鼠和人中發現在 ...
